Selecting Fluorescent Dyes for Long-Term Monitoring of Intracellular Calcium Influx Universitat Pompeu Fabra, España Intracellular calcium acts as a ubiquitous second messenger, regulating diverse cellular processes. We hypothesize that Tumor Treating Fields (TTFs) and other electric treatments may induce cytotoxicity primarily by disturbing calcium homeostasis via a mild form of electroporation. Intracellular calcium concentration is tightly regulated, maintaining a steep gradient between the cytosol (~100nM) and the extracellular environment (1–2 mM). This large difference drives calcium influx when the plasma membrane becomes permeabilized. Electroporation transiently disrupts membrane integrity, allowing extracellular calcium to enter the cell, rapidly altering calcium homeostasis and potentially leading to cytotoxicity. To study these dynamics, fluorescent calcium dyes are the most effective tools, as they bind free Ca2+ and emit fluorescence upon excitation, enabling real-time visualization of intracellular calcium changes with high spatial and temporal resolution via fluorescence microscopy. | |
